Alexander disease is a very rare and severe brain disorder that gets worse over time. It happens when a change in a specific gene causes the body to make too much of a protein called GFAP. This extra protein builds up inside brain cells and damages them.

Over time, this damage leads to trouble with walking, moving, swallowing, and thinking. Until now, care for people with this condition only focused on managing their symptoms. Zanvastro is designed to target the cause of the disease by lowering how much GFAP protein the body makes.

Why Was Zanvastro Approved?

The FDA approved Zanvastro based on results from a clinical study in 54 patients aged 1.5 to 53 years with Alexander disease. Most participants in the trial were children. The study compared patients who received Zanvastro infusions with a control group that did not receive the active drug.

Researchers measured walking ability after 61 weeks of treatment using a 10-meter walk test. Patients aged 5 and older who received Zanvastro maintained their walking speed, while the walking speed of patients in the control group dropped by about 35%. In younger children aged 2 to 4, motor skills improved in those taking Zanvastro compared to those in the control group. Most side effects were mild or moderate, and severe side effects happened less often in patients taking Zanvastro than in the control group.

What Do I Need to Know About Zanvastro?

Zanvastro is given as an injection directly into the spinal fluid, also known as a lumbar puncture or spinal tap. It is given by a healthcare provider once every three months.

Zanvastro can cause swelling or irritation around the brain and spinal cord, called aseptic meningitis. Tell your healthcare provider right away if you or your child develops symptoms such as a severe headache, fever, stiff neck, nausea, or sensitivity to light.

The most common side effects of Zanvastro include vomiting, back pain, cough, headache, joint pain, sore throat, difficulty swallowing, and pain or discomfort after a spinal tap. Tell your healthcare provider if you are pregnant, planning to become pregnant, or breastfeeding before starting treatment. Make sure your healthcare provider knows about all other medicines, over-the-counter drugs, and supplements you or your child take.

What Is Zanvastro and How Does It Work?
