Children with this condition lack an enzyme (sulfamidase) that is needed to break down a natural sugar complex in the body. Over time, this sugar builds up in brain cells and damages the central nervous system. This causes children to gradually lose skills such as talking, thinking, and moving.<\/p>\n<p>Fayuvi is a gene therapy given as a single infusion into a vein. It delivers a working copy of the missing gene directly into cells. This allows the body to make the missing enzyme and break down the sugar buildup. By restoring the enzyme, Fayuvi helps slow or prevent further brain damage in children who still have baseline learning and developmental abilities.<\/p>\n<\/section>\n<section>\n<h2>Why Was Fayuvi Approved?<\/h2>\n<p>The approval was based on a clinical trial that tested Fayuvi in young children with MPS IIIA. In the study, 17 children who received Fayuvi were compared to 27 untreated children who were not part of the study. Because Sanfilippo syndrome type A is extremely rare, researchers used this untreated \u201cnatural history\u201d control group to see how the disease usually progresses without treatment. The average age of the children who received treatment was 22 months, with ages ranging from 3 months to 33 months.<\/p>\n<\/section>\n<section>Researchers measured changes in thinking and learning skills between the ages of 2 and 5 years. Children treated with Fayuvi maintained or improved their cognitive abilities. In contrast, untreated children steadily lost those skills. On average, children who received Fayuvi scored 23.5 points higher on developmental tests than untreated children did. This difference showed a meaningful clinical benefit in preserving early childhood development.Safety was evaluated in a total of 33 children, including those from another small clinical study. Liver damage was the most common side effect, affecting 85% of children. Other common side effects included vomiting, fever, decreased appetite, and low white blood cell or platelet counts.<\/section>\n<\/div>\n<div>\n<section>\n<h2>What Do Patients and Caregivers Need to Know?<\/h2>\n<p>Fayuvi is administered as a one-time infusion into a vein. The infusion takes approximately one hour and is given in a specialized healthcare setting. On the day before the infusion, children must start taking an oral steroid medication, which will continue for at least eight weeks. Steroids help prevent liver inflammation and manage immune reactions caused by the gene therapy. Healthcare providers will order regular blood tests before and after treatment to monitor liver function and other blood counts.<\/p>\n<\/section>\n<section>The most common side effects of Fayuvi include increased liver enzymes, vomiting, fever, behavioral changes, diarrhea, decreased appetite, low white blood cell counts, and low platelet counts. Because low platelets can increase the risk of bleeding, caregivers should report any unexpected bruising or bleeding to a healthcare provider right away. Caregivers should also seek immediate medical care if the child shows signs of liver problems, such as yellowing of the skin or eyes.Patients should avoid receiving vaccines for 30 days before starting treatment and while taking steroid medications. Review the child\u2019s vaccination schedule with your healthcare provider before treatment.For three months after receiving Fayuvi, small amounts of the treatment vector can pass out of the child\u2019s body through bodily fluids and waste. During this time, caregivers should practice careful hand hygiene after touching bodily fluids or changing diapers.
