These terms describe specific proteins found on cancer cells that affect how the cancer grows. Revtorpyk is for people with this form of breast cancer whose cancer has come back or gotten worse after at least one hormone therapy.

The medicine is specifically approved for cancer that is locally advanced or metastatic. Locally advanced means it has spread to nearby tissue and cannot be removed with surgery. Metastatic means it has spread to other parts of the body. To qualify for Revtorpyk, your cancer must not have an abnormal change in a gene called PIK3CA. Your health care provider will test your cancer for this gene change to make sure the medicine is right for you.

Revtorpyk is a type of drug called a kinase inhibitor. It works by blocking specific proteins in your body that help cancer cells grow and multiply. By blocking these proteins, the drug helps slow down or stop the spread of the cancer. It is used in combination with a hormone drug called fulvestrant, and sometimes with another drug called palbociclib.

Why Was It Approved?

The FDA approved Revtorpyk based on a clinical study in 392 adults with advanced breast cancer. Researchers split the patients into three groups to compare how well different treatments worked. The first group (Group A) received Revtorpyk with fulvestrant and palbociclib. The second group (Group B) received Revtorpyk with fulvestrant alone. The third group (Group C) received only fulvestrant. The researchers measured how long each patient lived without their cancer growing or getting worse.

Patients in Group A, who got the three-drug combination, went an average of 9.3 months without their cancer growing. Group B (the two-drug combination) went an average of 7.4 months without cancer growth. Patients in Group C, who only got fulvestrant, went an average of only 2 months without growth.

The rate of side effects was higher in the two- and three-drug combination groups than in the group that only received fulvestrant. Some patients who received Revtorpyk had to stop treatment or lower their dose because of side effects like mouth sores, high blood sugar, or rash.

What Do I Need to Know About Taking This Medicine?

Revtorpyk is given to you by a health care provider as an infusion into a vein (IV). Each infusion takes about 30 minutes. The drug is usually given once a week for three weeks, followed by one week off. This four-week period is called a treatment cycle.

Mouth sores (stomatitis) are a very common side effect of this drug. These sores can be painful and make it hard to eat or drink. To help prevent mouth sores, your health care provider will prescribe a special steroid mouthwash that does not contain alcohol. You should use this mouthwash four times a day, starting on your first day of treatment. Do not eat or drink anything for at least 1 hour after using the mouthwash.

Revtorpyk can also cause high blood sugar. Your health care provider will test your blood sugar before you start and during your treatment. Tell your health care provider right away if you get symptoms of high blood sugar. These symptoms include feeling extremely thirsty, needing to urinate more often than usual, feeling very tired, or having blurry vision.

Skin rashes are another common side effect of this medicine. In some cases, these rashes can be very severe. You should limit your time in the sun to help reduce your risk of a serious skin rash. If you get a new rash, tell your health care provider right away.

This medicine can harm a fetus. If you are able to get pregnant, you will have a pregnancy test before you start treatment. Both women and men should use effective birth control during treatment and for two weeks after the last dose. You should also not breastfeed during treatment and for two weeks after your last dose.

The FDA has approved a new medicine called Revtorpyk (gedatolisib) to treat adults with a certain form of advanced breast cancer that is hormone receptor-positive (HR-positive) and HER2-negative.
